ABSTRACT

Objective:To explore the action mechanism of medicinal pair Astragali Radix-Angelicae Sinensis Radix against diabetic cardiomyopathy (DCM) based on network pharmacology and<italic> in vivo </italic>animal experiment. Method:The active ingredients and targets of Astragali Radix and Angelicae Sinensis Radix were retrieved from the Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform (TCMSP) and Bioinformatics Analysis Tool for Molecular mechANism of Traditional Chinese Medicine (BATMAN-TCM), and the relevant targets of DCM from the disease database. The common specific targets between the medicinal pair and DCM obtained via comparison were used for constructing the main active ingredients of Astragali Radix-Angelicae Sinensis Radix-DCM-target network, followed by protein-protein interaction (PPI) analysis of compound-DCM common targets and the screening of important modules using Molecular Complex Detection (MCODE) plugin. The gene ontology (GO) and Kyoto encyclopedia of genes and genomes (KEGG) enrichment analysis was carried out based on DAVID for exploring the possible molecular mechanisms, which were then subjected to experimental verification. Result:A total of 126 core targets of Astragali Radix-Angelicae Sinensis Radix for treating DCM were screened out based on network pharmacology. As indicated by KEGG pathway enrichment analysis, the above-mentioned key targets might be related to such pathways as inflammatory response, oxidative stress, insulin resistance, and apoptosis. The findings of <italic>in vivo</italic> animal experiments demonstrated that Astragali Radix-Angelicae Sinensis Radix delayed high glucose-induced cardiomyocyte injury of DCM rats, suggesting that this medicinal pair intervened in the pathological process to a certain extent. Conclusion:Astragali Radix-Angelicae Sinensis Radix alleviates DCM possibly by acting on multiple targets including interleukin-6, vascular endothelial growth factor A, tumor necrosis factor, TP53 gene, and nuclear transcription factor, regulating apoptosis and glucolipid metabolism, and improving oxidative stress and inflammatory response. The research based on network pharmacology and experimental verification has provided new ideas for investigating the pathogenesis of DCM and its clinical treatment.

ABSTRACT

Medicinal pair is the cornerstone in compatibility of traditional Chinese medicine prescriptions in clinical application.At present,there are many related studies on Chinese medicine for diabetic nephropathy,but they mainly focus on efficacy observation and prescription summary,lacking in-depth study and summary of the mechanism of action of traditional Chinese medicine (TCM).As compared with prescriptions,Chinese medicinal pair has fewer chemical components,a single form,simple mechanism of action,and fewer interference factors.In recent years,TCM has accumulated rich clinical practice experience in the treatment of diabetic nephropathy.Many researchers pay great attention to the application of medicinal pairs in the compatibility of prescriptions and their action rules.Through the compatibility of medicinal pairs,the optimized combinations can be achieved from various aspects to further improve the therapeutic effect of TCM compound.In order to clarify the ways and targets of Chinese medicinal pairs in treatment of diabetic nephropathy,relevant articles at home and abroad were systemically reviewed in this paper.For the clinical observation and animal experiments of the past 10 years,four types of medicines with "invigorating the spleen and the kidney" ,"clearing damp and promoting diuresis" "activating blood and removing stasis" and "securing and astringing" with curative effect for diabetic nephropathy,were mentioned in this article.The composition rules and the progress of experimental researches of these medicinal pairs were analyzed,while the compatibility principles,chemical compositions,and pharmacokinetic changesof the medicinal pairs were summarized to provide some references for their further application,researches and development.

ABSTRACT

Objective] To analyze the medication experience of Zhong Yi-tang for insomnia by using TCM inheritance support system 1.1ver.[Methods] The prescriptions of Zhong Yi-tang for insomnia were collected and input the data into TCM inheritance support system .The frequency and association rules of drugs were calculated by using data mining methods such as apfiofi algorithm,complex system entropy cluster and unsupervised hierarchical cluster,and medication rule of Zhong Yi-tang for epigastric pain was analyzed.[Results] Based on the analysis of 354 prescriptions, the frequency of each herb and association rules among the herbs were computed. Spine date seed, tuber fleeceflower stem, paeony, red sage root, boxthorn and Chinese Date were highly frequently found, 18 pairs of Chinese medicines based on these and 5 core combinations such as ganmai-dazao decoction were mined out . [Conclusion] Zhong Yi-tang usually used ganmai-dazao decoction, spine date seed, tuber fleeceflower stem and red sage root to cure insomnia. The sweet and moistening Chinese medcine were used to strengthen-spleen-stomach, benefiting ying-blood, and nourishing the heart-blood. Data mining method can be used to analyze old TCM doctors’clinical experience.
